Mechanism of Action

Melanotan I (MT1, also known as afamelanotide / Nle4-D-Phe7-alpha-MSH) is a synthetic analog of alpha-melanocyte-stimulating hormone (alpha-MSH) that binds melanocortin-1 receptors (MC1R) on melanocytes. MC1R activation increases intracellular cAMP and upregulates eumelanin synthesis via the MITF/tyrosinase pathway.

Structure and Class

A 13-amino-acid linear melanocortin peptide derived from alpha-MSH, modified with norleucine at position 4 and D-phenylalanine at position 7 to confer enhanced receptor affinity and metabolic stability versus native alpha-MSH. Class: melanocortin receptor agonist.

Primary Research Areas

Investigated in melanogenesis, photoprotection (erythropoietic protoporphyria), MC1R signaling, and pigmentary disorder models. Unlike Melanotan II, MT1 is more selective for MC1R and lacks the broader melanocortin receptor activity associated with MT2.

Notable Peer-Reviewed Findings

Levine et al. and subsequent investigators characterized MT1's MC1R-mediated melanogenic activity. Afamelanotide is approved in the EU and US for adult patients with erythropoietic protoporphyria, where clinical trials demonstrated reduced phototoxic reactions.
