Mechanism of Action
CJC-1295 (no DAC) is a modified growth hormone-releasing hormone (GHRH) analog that binds the GHRH receptor on pituitary somatotrophs to amplify pulsatile growth hormone release. Ipamorelin is a selective pentapeptide growth hormone secretagogue (GHS) that activates the ghrelin / GHS-R1a receptor without raising ACTH, cortisol, or prolactin. Co-administration provides a synergistic GHRH + GHS signal, producing a cleaner, more physiologic GH pulse than either peptide alone, with downstream increases in IGF-1.
Structure and Class
CJC-1295 (no DAC) is a 30-amino-acid tetrasubstituted GHRH(1-29) analog with a short circulating half-life (no drug-affinity complex). Ipamorelin is a synthetic pentapeptide (Aib-His-D-2-Nal-D-Phe-Lys-NH2) in the growth hormone secretagogue class. Both are lyophilized powders intended for subcutaneous reconstitution in bacteriostatic water.
Primary Research Areas
Studied in preclinical and clinical settings for endogenous GH/IGF-1 modulation, recovery and tissue repair models, sleep architecture, and age-related GH decline. The combination is widely referenced in educational dosing literature because the GHRH + GHS pairing magnifies pulse amplitude without flattening the underlying circadian rhythm.
Notable Peer-Reviewed Findings
Teichman et al. (2006) demonstrated sustained, dose-dependent GH and IGF-1 elevation with CJC-1295 analogs. Raun et al. (1998) showed Ipamorelin selectively releases GH without affecting ACTH or cortisol in rats and swine. Combined GHRH + GHS administration consistently produces a larger GH response than either agent alone in human and animal studies.
